ABSTRACT

We previously demonstrated that one of Rho small GTPase, Rac1, regulates canonical Wnt signaling and limb bud development through increasing the accumulation of beta-catenin in nuclei (Wu X. et al. Cell 2008;133:533-44), however, whether it is common effects of Rho small GTPase to regulate canonical Wnt signaling and limb bud development or not is still unknown. Base on our premier findings that another membrane of Rho small GTPase, CDC42, dose not regulate canonical Wnt signaling and canonical Wnt signaling-controlled limb bud development, but regulates endochondral ossification in conditional CDC42 knockout mice, we will further study the mechanism governing CDC42 regulates condensation, chondrocyte differentiation and hypertrophic differentiation in endochondral ossification, and study the potential upstream and downstream events of CDC42 signaling that regulates the cartilage development as well. This study will provide with the new theory for bone development that controlled by CDC42 and non-canonical Wnt signaling. More... »
