ABSTRACT

Exploring the mechanism of epilepsy formation and the ideal way to heal epilepsy is of great concern. It is known that endogenous neural stem cells in the hippocampal dentate gyrus granule are involved in the development of epilepsy abnormalities and implantation of exogenous neural stem cells can inhibit epileptic seizures, but how is the internal and external neural stem cells involved in the epilepsy Abnormal neurogenesis is not clear. This project is intended to demonstrate that hypoxia-inducible factor (HIF-1) regulates the involvement of internal and external neural stem cells in or affects epilepsy abnormalities in neurogenesis. Intratiplinary neural stem cells were labeled with BrdU in vivo. The exogenous neural stem cells were labeled with GFP and SPIO. MRI, electrophysiology, immunohistochemistry and electron microscopy were performed at the whole, tissue and cell level. And the expression of HIF-1α in hippocampus were measured to determine the effect of HIF-1 and exogenous neural stem cells on the occurrence of epilepsy abnormalities in the epileptic neural stem cells. The inhibition of HIF-1α expression by topotecan was further demonstrated by the correlation between HIF-1 and abnormal neurogenesis. On the basis of this study, the survival rate of transplanted neural stem cells and its efficacy is affected by HIF-1 regulation, for the treatment of epilepsy to provide new research ideas.
