ABSTRACT

Vancomycin-resistant enterococci (VRE) in recent years, the rapid increase in domestic separation rate. We found a novel gene cluster, vanM (AAC, 2010), which was resistant to vancomycin in the VRE strain. However, Southern hybridization showed a significant difference in vanM-positive plasmid size before and after metastasis. The mechanism of plasmid changes, the mechanism to be clear. In addition, the strain carrying the gene cluster has both VanA phenotype and VanB phenotype, and the mechanism of drug-resistant phenotype difference is to be studied. In this study, the following studies were carried out: (1) To understand the mechanism of drug-resistant gene cluster transmission by analyzing the sequence sequence of different vanM-positive plasmids before and after grafting; (2) To study the peripheral DNA sequence of vanM gene cluster carried by different resistant phenotype strains The relationship between the expression level and the change of the resistance phenotype was clarified, and the formation mechanism of different phenotypes was clarified. (3) The gene cluster and its expression product were screened by using gene mutation, deletion and protein spectrum analysis. Resistance-related key protein and functional domain, to find a new target.
