ABSTRACT

Colon cancer is a common malignant tumor of the digestive tract, seriously endangering people's health. Colon cancer cancer stem cells is the main reason for its development, targeting colon cancer cancer stem cell therapy is the current research focus. Our preliminary study found that the candidate marker of intestinal stem cells β1-integrin and Hedgehog (Hh) signaling pathway (tumor stem cell critical signal pathway) is closely related. On the basis of sorting out colon cancer stem cells (Lovo stem cells), β1-integrin was used to regulate the proliferation and apoptosis of Lovo stem cells through Hh signaling pathway. (1) Lentivirus-mediated gene silencing and overexpression were used to investigate the effect of β1- and the changes of Gli-1 promoter and other key members were observed. (2) After gene silencing, overexpression and antibody blocking of β1-integrin protein, the apoptosis and apoptosis of Lovo stem cells were observed at the cell and animal level. The aim of this study is to elucidate the mechanism of β1-integrin on Hh signaling pathway in Lovo stem cells, and to provide a theoretical basis for targeted treatment of cancer stem cells.
