ABSTRACT

ADAMTS-18 is a disintegrin metalloproteinase with thrombospodin motifs secreted by endothelial cells. Our previous studies revealed that a recombinant C-terminal fragment of ADAMTS-18 is capable of binding to the GPⅢa49-66 epitope of platelet, inducing platelet fragmentation by reactive oxygen species(peroxides) generated from a platelet NADPH oxidase pathway and 12-lipoxgenase, and destroying platelet aggregates. All of these data suggest that ADAMTS-18 could probably regulate platelet thrombus formation by inducing oxidative platelet fragmentation. In this study, we will verify the function of ADAMTS-18 in platelet fragmentation, arterial thrombosis and thrombolysis using an ADAMTS-18 gene knockout mouse model which will be established by ourselves firstly.
