ABSTRACT

Threaded potato virus X (PVX) is a typical member of poteksvirusov. For transport PVX genome from cell to cell requires three transport proteins (TB1,2,3) encoded in the so-called triple gene block and coat protein (CP). It is assumed that in the intercellular transport poteksvirusov engaged or fully assembled virions or nevirionnye complexes containing viral RNA, and BO TB1.Takie complex assembled in vitro, are transported from the cell-in-cells by microinjection experiments, but they still have the structure not been studied. In the project we will examine the conditions for assembly in vitro ternary complexes (PVX RNA-BO-Tupolev TB-1) describe the structure and assembly of products using atomic force microscopy (AFM), electron and immunoelectron microscopy (EM and PMM, respectively). Ability to RNA translation in the complexes will be studied in cell-free protein synthesis system from wheat germ (RFP). We use genetic engineering designs for the N- and C- terminal mutants Tupolev TB-1 and C-terminal mutants BO to study the role of individual functional domains of proteins in the formation of ternary complexes. We assume that the assembly soluble ternary complexes takes place in two stages. First, untranslated not fully dressed coat protein ( "odnohvostye") particles are formed by dressing protein 5'-terminal region of the PVX RNA. Then, Tupolev TB-1 molecules bind to the protein helix "head" of the particles, causing them to undergo remoduling and are broadcasted. Transportation form PVX may be not fully assembled viral particles or \ and fully assembled virions from the Tupolev TB-1, a protein associated with one end of the spiral. More... »
