ABSTRACT

Acute injury stress, high mobility group protein B1 (HMGB1) as a new advanced media participated in the uncontrolled inflammatory response and immune disorders pathological processes. Regulatory dendritic cells (DCreg) is a negative regulator of immune function DC subtypes play a crucial role in maintaining the immune balance. We recently discovered, HMGB1 stimulation associated DCreg immune response, but its exact mechanism is unknown receptor. The project first applied Toll-like receptor (TLR) 4 antibodies and TLR4-shRNA plasmids by antagonizing activation of TLR4 gene expression of TLR4 or down, observe the effect of blocking TLR4 receptors HMGB1 Effects DCreg differentiation and immune function; Secondly, TLR4 wild-type and mutant mice HMGB1 natural attack and severe burn model, to further explore the HMGB1 mediated by TLR4 receptor DCreg differentiation and immune effector mechanisms that may affect the argument after burn HMGB1-TLR4 pathway of functional status and its DCreg induced immune dysfunction in the body's important role pathophysiological significance for severe burns caused by complications of sepsis effective interventions provide new clues.
