ABSTRACT

Since the brain tumor stem cells may determine brain tumor development, invasion, metastasis, recurrence and whether or not the key to the treatment of sensitive cells. So traditional for all tumor cell populations should be replaced for the treatment of cancer stem cells, in theory, as long as the eradication of brain tumor initiating cells will be able to completely remove the tumor cells. The project is pleomorphic glioblastoma stem cells as an object, GBM isolated from cultured tumor stem cells, which express markers CD133 detected by immunocytochemistry and flow cytometry, screened after CD133-positive cells were seeded to SCID mice produced a mouse model of GBM brain. The temozolomide (TMZ) made of nanoparticles with anti-CD133-specific monoclonal antibody crosslinking, the crosslinked TMZ -Ab - NPs direct effect on tumor stem cells in order to observe the killing effect of the molecule targeted therapy for brain tumor stem cells in vivo experiments will TMZ -Ab - NPs injected into the SCID mouse GBM, then observed for GBM by the cultured tumor stem cells caused by the therapeutic value of such antibodies conjugated TMA nanoparticles.
