ABSTRACT

Using different substitution lines and synthetic hexaploid wheat lines bred by our group as the main experimental materials to construct U by high performance capillary electrophoresis, two-dimensional electrophoresis, mass spectrometry and biological differences in fluorescence two-dimensional electrophoresis (DIGE) and other proteomics, C , S, D and other relatives of the wheat genome encodes a protein expression profile, combined with the quality parameter analysis to identify candidate proteins subunits and quality-related, including LMW, gliadin, clear globulin; cloning quality-related proteins coding genes, analysis of gene molecular structure and allelic variability of bioinformatics tools to predict gene function; build an efficient expression vector and high-quality candidate genes in E. coli, the expression in vitro using purified protein recovered trace Blending method further verification gene function; according to variability of cloning candidate gene SNPs, InDels, focusing on screening PCR-based marker gene functions, and through marker-assisted selection to verify their reliability; studies of candidate genes from high-level expression of transcriptional and translational regulation of its relationship with relationship quality formation; use methods such as Network build a different genome phylogenetic tree wheat systems and differentiation timetable for further study of the origin and evolution of the wheat genome of cultivated wheat.
