ABSTRACT

The goal of this project was to study the effects of chronic calcium antagonist treatment on the electrophysiological and molecular biological parameters of cardiac muscle. Dogs were treated with calcium channel antagonists for 4 weeks. The serum nifedipine concentration the ECG parameters and the blood pressure values were monitored continuously. The chronic nifedipine treatment significantly reduced the calcium current (ICa-L) amplitude by 48 ± 13% in all the layer of the left ventricular wall, but it had no effect on the amplitude and the kinetic parameters of the potassium currents(Ito, IKr, IK1). In the cardiac tissue samples the amount of calcium channel alpha subunit was significantly less after the chronic calcium antagonist treatment than that in control. The immunofluorescence labeling was also reduced by nifedipine treatment. We could not detect any change in the diastolic interval dependency of the action potential duration (APD50 and APD90) after the 4 weeks treatment. The chronic calcium antagonist treatment had no effect on he amplitude of intracellular calcium transients, but it reduced the resting calcium concentration and speeded up the process of calcium elimination.
